Abstract/Summary:
Vitis davidii Foex in Luocheng is a species of Chinese wild vitis . In this paper,the in vitro culture of its single bud with stem segment has been studied,inducing callus and differentiation of its leaves have also been studied . The results were:(一)stem culture1 Using 0.1% Mercuric chloride was the better method for explant sterilizing.2 Taking the breaking bud from dormant branches which were sand cultured in room was the best way of getting explants .3 Either 1/2MS or B5 medium was sutibale for culture .4 It is not suitable to use 6-BA and KT in tissue culture of Vitis davidii Foex, because they should cause explant browing and dying.5 ZT1.0-2.0 mg/L with low concentration of NAA about 0.01-0.05 mg/L was the best match to breaking the axillary bud for primary culture .6 B5+ZT1.0 mg/L+IAA0.01 mg/L was the best medium for subculture , the plantlets grow well ,and the multiplication was highest ,getting 2.01.7 It wae easy to get root from the plantlets.1/2MS+IBA0.5 mg/L was the most suitable medium for rooting . The rooting rate got 100%.(二) Inducing callus and differentiation of its leaves1 It was very difficuilt to get regeneration plant from its leaves ,no regeneration plant had been differentiated in this experiment.2 Using auxin NAA,IBA or cytokinin ZT alone could induce callus, but if using ZT and NAA together should be better ,1/2B5+ZT1.0 mg/L +NAA0.01 mg/L was the most suitable medium for inducing callus from leaves, the inducing rate got 100%,and the callus was compact, granulated. Adventitious root was easy to be differentiated from leaves when use NAA or IBA, the rooting rate was getting higher when the concentration of auxin getting higher .3 ZT 1.0-2.0 mg/L with low concentration of auxin (0.005-0.05 mg/L) was a suitable match for callus subculture , it could make the callus keeping compact , granulated , and energetic . In primary culture, this cultivar was easily hurt by disinfector, and its stem disjoint frequently, then, the survival rate was low. This cultivar was one of narrow adaptability species. It was easy to induce callus, so,the strict medium was requested, then the proliferous multiple could catch the aim. It was need to do more research.
