In order to research the effects of sleep deprivation(SD) on the pubertal rats,the model of REM SD rat was established by using plat form method.Immunohistochemical ultrasensitive SP method was used to investigate the expression of NGF and Ob-RL in the brain,pituitary and testis of pubertal rats.The main results are as follow:1,The expression of Ob-RL in the hippocampus was decreased after 1d to 3d SD and increased after 5d SD. In the hippocampus,leptin is related to the study and memory,the abnormal expression of Ob-RL is related to the decrease of learning and memory ability,the increased expression of Ob-RL after 5d SD is related to the oxygen deficit after long time SD。The expression of NGF in the hippocampus was increased gradually as the time of SD increased. SD can demage the neurons or make the neurons apoptosis in hippocampus,the increasing expression of NGF may be a self-protection of the brain.2,In the auditory nucelus,the expression of Ob-RL after SD is just like in the hippocampus. The expression of Ob-RL was decreased after 1d to 3d SD and increased after 5d SD. The decrease expression of Ob-RL in the auditory nucleus can make the expression of 5-HT increase which is related to the tinnitus and acousma.In the DC,VC,MMGN and CMGN,1d to 3d SD can make the expression the NGF increase temporarily.As the increase of SD time,the expression of NGF began to decrease.In SOC,VLL,ECIC,CIC and AC,the expression of NGF is decrease as the SD going on.In the DLL,the effect of SD on the expression of NGF is not significant. NGF is close related to the earlier development of efferent nerve fibers in the auditory system and can protect auditory neurons from acquired pathological changes.otherwise,it can promote the survive and regeneration of cultural auditory neurons.the decrease of the expression of NGF in the auditory nucleus may affect the normal function of auditory system.3,In the hypothalamus, the expression of NGF and Ob-RL is decrease as the SD going on.The decrease of expression of Ob-RL is close related to the increase of food consumption of rats after SD,it may also lead to the decrease of testosterone and GH which can affect the development of reproductive system and body growth. the decrease expression of NGF after SD may have the same mechanism of the decrease expression in the ischemia-reperfusion brain.the decrease expression of NGF can affect normal function of endocrine neurons and affect the endocrine system.In the anterior pituitary, the expression of Ob-RL was decreased after 1d to 3dSD and increased after 5d SD which may affect the normal secretion of GH,LH and FSH. The expression of NGF after SD is decrease which may reduce the mature PRL cells and restrain the differentiation of PRL cells.In the testis, the expression of Ob-RL is close related to the spermatogeny cycle. The expression of NGF and Ob-RL is increase as the SD going on. The increase of Ob-RL in the testis can promote the of the sensitivity of leptin, thus restrain the secretion of testosterone which affect the development of male reproductive system.
