As an initiative central nervous system adjustment,sleep is a complex physiological process in human natural circadian rhythm.It plays a critical role in growth,development,learning and memory.As we know,the fast-pace life has made sleep deprivation(SD) more popular and influenced the daily life seriously.Adult male rats were subjected to 24,48 and 72h SD using single platform methods.The mRNA expression level of c-fos,nestin,bid3,jak2in the hippocampus,cerebellum,and cortex of SD rats was identified by RT-PCR.The change of protein level was also detected by immunohistochemistry.These studies would promote the further research of the molecule mechanism of physiological changes caused by sleep deprivation.Part one:mRNA expression of c-fos,nestin,bid3 and jak2 in SD rats.Methods:REM sleep deprivation was induced by housing rats on the small platform over water.Control groups were housed in the normal cage(C) or on the large platform(LP) over water.The mRNA level of c-fos,nestin,bid3 and jak2in hippocampus,cerebellum and cortex was examined at24,48 and 72h during sleep deprivation by RT-PCR.Results:The expression level of c-fos,bid3 and jak2 was up-regulated in SD groups,and differences existed between the SD and LP groups;nestin was down-regulated in SD groups compared with control. The significant difference appeared in the hippocampus.Part two:Protein expression of Fos,Nestin,Bid3 and Jak2 in SD rats.Methods:REM sleep deprivation was induced by housing rats on the small platforms over water.Control groups(C) were housed in the normal cage. The expression level of Fos,Nestin,Bid3 and Jak2 in hippocampus,cerebellum and cortex was examined at 24,48 and 72h during sleep deprivation by immunohistochemistry.Results:Compare with the control group(C),Fos,Bid3 and Jak2 protein expression was increased at hippocampus,cortex and cerebellum in SD group.Our studay showed that the mRNA and protein level of c-fos,nestin,bid3 and jak2 were both regulated in rat CNS during sleep deprivation.Sleep deprivation could up-regulate c-fos;bid3 and jak2 mRNA and protein level,and down-regulate nestin expression level.Meanwhile,the expression level of same gene is inconsistent in different parts of the brain.Even in the same part of the brain,the change of mRNA and protein level shows different expression profiling.The reason is probably that different parts of the brain response differently to sleep deprivation and the same gene play different role in different central nervous system cells.
