Aldehyde dehydrogenase 2(ALDH2)is the key enzyme involved in acetaldehyde metabolism in human body, which can catalyze a variety of aldehydes into corresponding carboxylic acids including aliphatic aldehydes and aromatic aldehydes and prevent poisonous attack from aldehydes in the cells. ALDH2 gene has single nucleotide polymorphisms whose genotyping and frequencies of the polymorphisms exist differences among the individuals and also races. Among asians, there are high frequent mutant types allele ALDH2*2 gene. The inherited deficiency of ALDH2 results in the differences in the alcohol metabolism among alcohol drinkers, which affects humans drinking behavior, the intoxicated degree and organ demage induced by alcohol and so on. In recent years, the research on ALDH2 polymorphisms draws more attentions, and people expect to find out a method in order to the detect the genotyping rapidly and discuss the relationship between genotyping of ALDH2 and the disease resulting from alcohol drinking. Hopefully, it can offer the credible data for the relevant research, and accordingly can prevent and decrease the occurrence of the disease.In this paper,genome DNA was extracted by two methods, which were one-step method and mouthwash method, respectively. 127 random samples were under the genotyping detection by PCR-RFLP and AS-PCR and the frequency of these three kinds of genotyping were analyzed statistically.The results showed that the genome DNA can be extracted rapidly by the mouthwash method. The results of genotyping detection among 127samples were in coincidence through PCR-RFLP and AS-PCR, and also the distribution of genotyping frequency accorded with the Hardy-Weinberg equilibrium. In addition, it was concluded that AS-PCR is a kind of method which can detect the gene polymorphisms more simply and more rapidly.
