Marked on cytochrome C oxidase I(COI) gene part sequence (914bp) revealed23species of Gruiformes distribution in China,we reconstructed the phylogeny and molecular evolution relationship of Gruiformes.Based on maximum parsimony(MP) and Maximum likelihood(ML)analysis implemented in paup*with stepwise addition of the starting tree,all species in Gruiformes showed polyphyletic. Especially,Turnix tanki did not group into traditional Gruiforme, but into the clade of Charadriiforme. This paper also ensured that Hydrophasianus chirurgus belonged to clade of Charadriiforme.Our molecular taxonomy on the Amaurornis was different to the traditional, the closer relationships were detected between Amaurornis akool and Porzana and between Amaurornis phoenicurus and Gallicrex, respectively.We also found that Gallinula was a sister group to the clade of Fulica with strong support.On the other hand,analyzed on mitochondrial DNA control region part sequences (612bp) from72specimens in Porzana fusca,we found22different haplotypes, within mainlandof China, of which11were geographical population. Except for the Fuan populations,there was not significant divergence among populations in all geographical population(Fst=0.07378;P-value=0.06549).Gene flow was higher among populations.On AMOVA analyzed,population genetic diversity was shallow between all the geographical populations, indicating range expansion with high within populations exchange(92.62%within populations) and with low among populations exchange(7.38%among populations). This study provided support for the Pleistocene of last postglacial expansion hypothesis in Porzana fusca in South China and suggested that postglacial expansion a population contraction occurred, which would tend to erase less frequent haplotypes, and then the population expanded in size on Mismatch analysis. Neutrality test and Mismatch analysis showed Fuan,Ruijin,Changting populations expanded in size.There was no obvious correlation between gengtic distance and geographic distance in different populations by Mantel test(R=0.1515,P=0.2280).
