Nicotine was a main alkaloid and toxic compound in breeds of tobacco, which was one of environment contamination. Some microorganisms were able to utilize nicotine in different pathway, some of which could be applied in the tobacco waste treatment. Only a few studies about nicotine-biodegradation could be found in home.In this paper a novel nicotine-degrading bacterium named ZUTSKD was isolated from waste tobacco. On the basis of morphological, physiological charaters, Biolog GN, 16S rDNA sequence and phylogenetic characteristics, strain ZUTSKD was identified as a member of Pseudomonas sp.Nicotine could be utilized as a sole source of carbon, nitrogen and energy by Pseudomonas sp. ZUTSKD. The optimal condition for nicotine degradation was 30℃with initial pH 7.5. The addition of NH4Cl and glucose in the medium could improve the nicotine degradation by Pseudomonas sp. ZUTSKD, but the addition of sucrose could not. This paper showed that Pseudomonas sp. ZUTSKD had high ability to degrade nicotine in tobacco waste and Extracted liquid of tobacco. Through domesticating, domesticated strain of Pseudomonas sp. ZUTSKD could degrade nicotine in high concentration tobacco waste. Pseudomonas sp. ZUTSKD would be one potential strain that could be directly applied in the tobacco waste treatment and tobacco production process.Four new intermediate metabolites such as 3-(3,4-dihydro-2H-pyrrol -5-yl)-pyridine, 2,3'-bipyridine, cotinine and 3-pyridinecarboxylic acid could be detected in the medium by GC-MS. It was concluded that Pseudomonas sp. ZUTSKD had a new pathway of nicotine degradation possibly.
