A preliminary observation on the early embryo of A.igrocauda was made use of artificial propagation,dry fertilization and embryo fixation.This research carried on WISH using model organism—zebrafish's two genes:nanos and eomes and different stage embryos of A.nigrocauda to study the two genes' expression during A.nigrocauda embryonic development,the results as following:The embryonic development of A.nigrocauda needs about 50 h from fertilization to hatching at 24℃,it experiences cleavage,blastocyst,original intestinal period,organ development and improve t,and then hatching.The embryo is overall transparency except for eyespots after hatching.The nanos as a marker gene for germ cell expresses in the primordial germ cells (PGCs).The RNA of nanos can be checked as early as about 4 h after fertilization,at first its expression is scattered,then concentrated and migrates to reproductive ridge.The expression of nanos during early embryonic development of A.nigrocauda is different from that in zebrafish during the same stages,then its expressions gradually concentrate and different,showing a certain expression pattern.The eomes is a marker gene that expresses in telencephalon.Its RNA expression starts as early as 7 h after fertilization and strengthens in the later stages,then migrates to two sides symmetrically.During A.nigrocauda early embryonic development,it was observed in the different expression from that in the zebrafish.The eomes's expression patterns were the same as that in zebrafish during the latter stages.There are at least two key aspects that it is necessary to be attented using WISH to analyse gene expression patterns between different species:1:The similarity between the sequences of genes in defferent spacies2:Gene expression pattern,such as position and productionThe genge that with high sequence similarity and expression quantity,and also surface expression can be used WISH to study its expression in another spacies.
