Signal transduction and transcription activated factor(STAT)inhibitory factor(PIAS)can interact with many proteins,PIAS protein can influence the activity and function of other protein,and participate in the gene regulation of the ranscription as well as the cell signaling pathways,have important physiological function.PIAS protein family control mechanism basically has two kinds:one kind is by its own SUMO(small ubiquitin-related modifiers)E3 connection enzyme activity,promote chemically modified to some transcription factors and auxiliary transcription factor,especially the SUMO decoration,thus regulate the transcription activity;The other is as structure protein to provide the platform for protein-protein interactions,and promote cell signaling pathways gene transcription complex compounds or other regulatory protein removal and raise,and involves the target protein of the orientation of the nuclear.In order to study the PIAS1 related functions in mammalswe get the model animals zebrafish for study its embryogenesis period and the development of organization analyzing the mRNA and protein expression level.RT-PCR results show that PIAS1 genes in the embryos and organization are widespread expressed,but there exists express differences,mRNA expression began to increase at the gastrula stage,as it is the highest period in the preembryonic development,at this time all sorts of relevant factors should start a lot expression in vivo,and at the opticrudiment stage have a large expression,we see its transcription and translation by many factors and then restrain and promote its expression;Western Blot results show that PIAS1 protein in zebrafish embryos and organizations have universal expression,but have difference,for the protein synthesis affected by posttranslational modification and interaction with other cell factors to strengthen and inhibit expression,we see the expression in the organization of the ovary and testis have big differences among other tissues,we conjecture it might be involved in spermatogenesis and eggs generation.In order to further research PIAS1 genes function,we construct pEGFP-PIAS1 antisense vector,injected in the zebrafish single phase through the development of embryos,and the results showed that the overexpression of the PIAS1 have 80%of apoptosis or growth unusual in embryo development,reveals PIAS1 plays an important role in the process of zebrafish embryonic development and organization differentiation.
