P. fulvidraco was studied in this study. Its snout,tail fin,kidney,heart were cultured by tissue explant,in 27℃,with the medium 199(10 %FBS). The results showed that the snout cells migrated from the tissues after cultured in vitro for 3~5 h, it was earlier than other kinds of tissues. The snout cells have been subcultured to the eighth generation by trypsin digestion method, and tail fin cells to the forth generation. The cells of snout and fail fin were both fiber-like cells. The cells of kidney were like epithelial cells, while the cells of heart were like fibroblasts. When cultured under different temperature (15,27,37℃) , serum concentration (0,10 %,20 %), differnt medium (M199,M1640,MEM),snout cells were most quickly multiplied under 27℃,10 % serum concentration,M199, which could be seen from the growth curve of the snout cells. The appearance of cells was observed by H33342 and H.E. The normal cell nucleus included dyeing light colour, clear strucure, smooth nuclear envelope and average chromatin spot by H33342. Inversely,the apoptotic cells included chromatin condensation and aggregation at the periphery of the nucleons and nuclear fragmentation. Snouts and tail fin cells multiplied rapidly in the logarithmic growth phase by H.E.The slides were prepared according to the routine chromosome preparation technique with snout tissues and cells. They were observed under the microscope and it was found that the number of chromosomes were the same. The snouts cells have 2n=52 chromosomes, accounting for 71.55 % of the cells, of which was NF=90 and the karyotype formula was 2n=26m+20sm+6t. The chromosomes were normal.
