This paper studied Coriolus versicolor for the effect of some metal ions and inductors as well as co-culture of Coriolus versicolor with filamentous fungi to the secretion of laccase. Also we established fed-batch and pH regulation technique and appropriate fermentation conditions for Coriolus versicolor in order to improve laccase production and reduce production costs. The results are as follows:(1) Add Cu2+ with appropriate concentration of 300μmol/L after inoculation 2d, laccase activity and the content of extracellular protein reached 4558.4U/L and 2.65mg/ml. It was more appropriate to add 500μmol/L Mn2+ after inoculation 3d.(2) The appropriate concentration of vanillin and guaiacol were 250μmol/L and 20μmol/L. Two self-made, cheap and convenient small molecule medium A and B as inducers can effectively promote the synthesis of laccase. Laccase activity reached 3493U/L using the small molecule medium A as inducer.(3) The optimum initial concentration of wheat bran and the pH value were 20g/L and 5.0. pH regulation and fed-batch fermentation were of great advantage for laccase secretion by Coriolus versicolor. The maximum activity reached 3366.7U/L and extracellular protein content was also significantly increased using 20g/L wheat bran as nutrition feeded for three times during the fermentation. When the fed amount was constant, there were three approaches of fed-batch as increasing, constant, decreasing, during which the best way of fed-batch was the decreasing mode.(4) For co-culture of Coriolus Versicolor and filamentous fungi in liquid fermentation, the appropriate time for inoculation was 4d of the culture of Coriolus versicolor, laccase activity was 2.2 times of the control.
