Objective:The present thesis designed an ovariectomized rat model, through the exercise and estrogen replacement therapy, to explore the effects of exercise and estrogen on PPARγand GSK-3β, P-GSK-3βof lipid metabolism in ovariectomized rats.Methods:The ovariectomized female Sprague-Dawley rats, provided exercise and estrogen replacement for a period of 15 weeks, were randomly divided into five groups:Sham, Sham+E, OVX, OVX+E and OVX+E2. The exercise groups would performed 18m/min, gradient of 5 degree,45min and five days a week; The OVX+E2 group was injected the estrogen according to the weight 25ug/kg weekly, three times a week. After the experiment, measured the intra-abdominal fat mass in rats, and measured serum levels of estrogen(E2) by radio-immunitied. The fat cell morphology would be measured by HE staining. The total protein content and adipose tissue PPARγ, GSK-3βand P-GSK-3βprotein were determined through Bradford and Western blot.Results:(1) After 15-week experiment, the body weight in OVX was significantly higher than Sham (P<0.05), and after exercise and estrogen replacement therapy it respectively decreased(P<0.05)(2)after ovariectomized, the intra-abdominal fat OVX rats were heaviest in all group, and through exercise and estrogen replacement, fat mass were decreased significantly (P<0.05) (3) the triglyceride content in OVX adipose tissue was significantly higher (P<0.05), after exercise and estrogen replacement therapy the triglyceride significantly decreased (P<0.05) (4) the OVX area of fat cells significantly increased (P<0.05), through exercise and estrogen it was significantly reduced (P<0.05) (5) compared with Sham, the Ovx rats PPARγexpression was increased, after exercise, Sham+E and OVX+E adipose tissue PPARγexpression were increased, and E2 can also increase the PPARγexpression(6)the exercise group P-GSK/GSK protein expression in adipose tissue were significantly higher than quiet group (P<0.05); OVX+E2 group protein expression in rats no significant changes (P> 0.05). OVX rats protein expression in adipose tissue P-GSK/GSK slightly lower than the Sham. Conclusions:(1) Exercise and estrogen replacement therapy in vitro, can certain release the ovariectomized rat intra-abdominal fat accumulation (2) Moderate-intensity exercise can improve the ovariectomized rats estrogen (E2) levels (3) Exercise increase adipose tissue PPARγ, P-GSK-3β/GSK-3βprotein expression, promote the adipose tissue metabolism;estrogen can also increase adipose tissue PPARγprotein expression, and no influenced on P-GSK-3β/GSK-3βprotein expression.
