Hypothalamic-pituitary-gonad axis (H-P-G axis) is a very important neuroendocrine regulation axis in organism, the regulation function of HPG axis mainly relies on interaction between the information objects--Hormone, cytokine and neurotransmitter what exist in organism. As the reseach indicates, cytokine influences reproductive neuroendocrine, the function of ovary, the development and function of uterus and etc., but hormone influences the synthesis and secretion of cytokine besides regulates the development and function of reproduction system.This study was designed to know the cellular mechanism of estrogen, estrogen cooperative cytokine (interferon-γ, IFN-γ) on uterine protection, using the method of immuno-histochemical SP to investigate the IFN-γ, Bcl-2 and Bax positive expressions in uterus of ovariectomized rats before administration and 2, 4, 6, 8 weeks after administration by respectively injecting 17-βestrodiol, 17-βestrodiol combined with three different dosages IFN-γto four ovariectomized model groups. The major results were as follow:1. The effects of 17-βestrodiol cooperate IFN-γon the expression of IFN-γin uterusIFN-γimmunohistochemical positive substances were located in uterine endometrium of rats and main located in cellular membrane, cytoplasm, and weren't located in uterine muscular and serosa. After ovariectomized, relative expressions of IFN-γpositive productions(μ2) tapered with the administration course and to be the minimum when administering to 6 weeks, which had significant difference with the SHAM group(P<0.05),theμ2 of IFN-γhad a little increase after administer 6 weeks and still had significant difference with the SHAM group(P<0.05). After treated with 17-βestrogen, theμ2 of IFN-γincreased and had remarkable difference with OVX group(P<0.01), but theμ2 of IFN-γmaintain the level of 6 weeks after administer 6 weeks and had no significant difference with the SHAM group. After treated with 17-βestrogen cooperate three different IFN-γdoses, theμ2 of IFN-γhad keep increase with the administration course, which had very significant difference with OVX group(P<0.01), and had significant difference with OVX+E2 group(P<0.05),but had no remarkable difference with SHAM group.2. The effects of 17-βestrodiol cooperate IFN-γon the expression of Bcl-2 and Bax in uterus Bcl-2 and Bax immunohistochemical positive substances were located in uterine endometrium and muscular, which weren't located in uterine serosa. Bcl-2 was mainly located in cellular membrane, cytoplasm and Bax was located in cellular nucleus besides cellular membrane, cytoplasm. After ovariectomized, theμ2 of Bcl-2 tapered with the administration course and to be the minimum, concurrently, theμ2 of Bax increased, and to be the maximum, which had significant difference with the SHAM group(P<0.05). After treated with 17-βestrogen, theμ2 of Bcl-2 increased and had remarkable difference with OVX group(P<0.05), and theμ2 of Bax decreased and had significant difference with OVX group(P<0.05) too. After treated with 17-βestrogen cooperate 50IU or 200 IU IFN-γdoses, theμ2 of Bcl-2 had very significant difference with OVX group(P<0.01), and theμ2 of Bax decreased and had very significant difference with OVX group(P<0.01), but they had no remarkble difference with SHAM group. After treated with 17-βestrogen cooperate 800 IU IFN-γdose, theμ2 of Bcl-2 had significant increased, and theμ2 of Bax had significant decreased at the administer 0-4 weeks compared with the group of treated with 17-βestrogen cooperate 50IU or 200 IU IFN-γdoses, the apoptosis of uterus had no significant different after 4 weeks, and had no significant difference with the group of treated with 17-βestrogen.All the results illustrated that the body had a regulation mechanism can improve immune function of uterus in a certain degree as endogenous estrogen deficiency in organism. estrogen in proper dose had certain protective effect on uterus in the all stage of endogenous estrogen deficiency, 17-βestrodiol cooperative IFN-γin proper dose influences the synthesis, secretion and cooperation of cytokine in HPG axis.
