Protein is a fundamental and important component of the organism. Accordingly, the researches on protein are essential to recognize the essence of the organism. However, there are thousands of proteins in nature. It has been focused on by the scientists how to separate and purify these proteins. Recently, molecular imprinting technique has been introduced to this field with the development of the protein separation technique. In this work, chitosan and its derivative were taken as the imprinting material, and two kinds of protein imprinting polymer were prepared by different methods.A novel protein imprinted polymer was prepared by LBL self-assembly technique and molecular imprinting technique. The negatively charged bovine serum albumin and positively charged chitosan were alternately deposited onto the surface of the crosslinked chitosan substrate. After crosslinked by epoxy chloropropane and eluted by 5%SDS-2%HAc solution, the protein imprinted polymer (L-CS) was prepared. The results showed that the surface of the imprinted polymer became rough, and the water contact angle of the surface evidently decreased by 70%. The BSA adsorption capacity of the imprinted polymer was 13.67 mg/g, which was 27.7 times that of the non-imprinted polymer. The protein imprinted polymer had the selective adsorption ability towards the template protein BSA.The carboxymethylated chitosan (CMCS) has been synthesized. Then, a protein imprinted polymer (CS/CMCS-MIP) was prepared by using the chitosan and carboxymethylated chitosan compound as functional matrix, bovine serum albumin as template molecule, and epoxy chloropropane as the crosslinker. Protein adsorption experiment showed that the template protein adsorption capacity was 30.8 times higher than that of the non-imprinted polymer. CS/CMCS-MIP also showed the selective adsorption ability towards the template protein BSA.In this work, chitosan and its derivative were selected as the protein imprinting material. It can provide important theory and application data for preparing the material on protein selective adsorption and separation.
