PCSK5 is a member of proprotein convertase family. A wide vartiety of proteins are synthesized as inactive precursors that are subsequently converted to their mature active form by PCs. It's shown that PCSK5 can proteolysis some growth factors and receptors, for example epidermal growth factor and activin A. It's shown that those growth factors play an important role in mice implantation It's suggest that PCSK5 may involved in embryo implantation. There is still no report about function of PCSK5 in mouse implantation.The aim of this study was to investigate PCSK5 expression in the mouse uterus during early pregnancy, pseudopregnancy, delayed implantation and activation, artificial decidualization, steroid hormone treatment by in situ hybridization, and to analysis PCSK5 mRNA expression in decidualization uterine stromal in vitro by RT-PCR. PCSK5 mRNA was not detected from days 1 to 4 of pregnancy. When embryo implanted on day 5, PCSK5 mRNA were strongly detected in the sub-luminal stroma immediately surrounding the implanted blastocyst. There was a high level of PCSK5 expression in the primary decidual zone on days 6 to 8 of pregnancy. It's shown that PCSK5 may be involved in embryo implantation and decidualization of stromal cells. PCSK5 mRNA was not detected in mice uterus of pseudopregnancy and delayed implantation, but in uterus of activation delayed implantation, PCSK5 was showed a very similar distribution compared to embryo implanted on day 5. It's shown that expression pattern of PCSK5 is implantation-specific expression and PCSK5 expression is induced by the active blastocyst. PCSK5 was exited in the primary decidual zone of artificially induced decidua uterus. The date from decidualization uterine stromal cells induced by hormone in vitro, it's shown that PCSK5 mRNA gradually increases in the decidualization processing. It suggests that PCSK5 may be involved in decidualization of stromal cells.
