The heterocyclic derivatives of pyrimidine have become critical intermediates for drugs, pesticides and organic synthesis. The optimization and improvement of pyrimidine synthesis will greatly promote the development of related industries.In this dissertation,4,6-dimethylpyrimidine was synthesized via one-step reaction, replacing the original two-step reaction which was obtained by the reduction of 2-mercapto-4,6-dimethylpyrimidine with Raney nickel. By optimizing the purification procedure, the purity of the final product can reach 99%, much higher than the commercial available from Aldrich company.Additinally,2,4-dichloro-6-methyl-5-nitropyrimidine was synthesized with the 6-methyl-1H-pyrimidine-2,4-dione as the starting material, through the two steps of nitration and chlorination. It is easily purified with vaccum distillation instead of recrystallization method.The two pyrimidine derivatives are very important pharmaceutical intermediates with bringing good economic benefits due to their high commercial value.
