In the dissertation, we summarized the characteristics and the latest development of cubane, homocubane and their derivatives. Their application in pharmaceutical synthesis was also mentioned. A series of homocubane amides and esters have been synthesized and their biological activity has been evaluated. On the basis of reports on homocubane compounds, we chose the best route to prepare homocubane-2,4-diacid. Then ten of its amides and six esters have been synthesized. They are homocubane-2,4-(N-(4-benoxyl) phenyl) diamide (TML); ho m o cub an e -2 , 4- (N - (3 , 5 - di fl u or o methyl) ph en yl) di am i d e (TM 2); ho m o cubane-2,4-(N-(ethylacetate)phenyl)diamide(TM3 );homocubane-2 ,4- (N-(5 -nitro-2-amino)piperidyl)diamide(TMS);homocubane-2,4-(N-(2 -chloro-3 -amino)piperdyl)diamide(TM6);homocubane-2 ,4-(N-(6-met hyl-2-amino)piperdyl)diamide(Tm7) ;homocubane-2,4-(N-morpholine )diamide(TM 10) ;homocubane-2,4-(N-(5 -nitro-2-amino)thiazolyl)dia mide(TM 11) ;homocubane-2,4-(N-(2-amino- 1,3 ,4-thiadiazole)diamide (TM 12) ;homocubane-2,4-(N-(5 -amino)tetrazole)diamide(TM 13) ;hom ocubane-2 ,4-(phenol)diester(TM 14) ;homocubane-2,4-(4-nitro-phenol )diester(TM!5); homocubane-2,4-(4-chloro-3-methyl-phenol) diester (TM 16) ;homocubane-2 ,4-(4 -chloro-3 ,5 -dimethyl-phenol)diester(TM 7);homocubane-2,4-(4-acetamido-phenol)diester(TM 1 S);homocubane -2,4-(4-phenyl-phenol) diester (TM 19). The work is the beginning of studies on homocubane derivatives as biomedical agents.It has created opportunities for pharmaceutical development in the future.
