The recombinant strain harboring the carotenoids biosynthesis gene was constructed via genetic engineering, and two recombinant strains (R^ and R^) were selected. An analysis of carotenoids production was studied. The results showed:(1) The optimum fermentation conditions were as follows:Medium of 1% molasses, firstly it grew at 37C on shaker for 9 hours, then IPTG was added and cultivated at 23 C for 39 hours.(2) An optimum method of carotenoids extracted from recombinant strain had been confirmed. The cells were immersed in 6ml oGmol/L HCI on shaker at room temperature for 60 min, and were taken into burning water for 4 rain, then were cooled quickly and centrifugated, at last extracted by acetone.(3) The pigment was separated by thin-layer chromatography on kiesel gel plate with methanol : acetone : ligarine = 2: 5 : 3. The carotenoids were composed of at least five components.(4) Fermentation were conducted in 1L and 3L fermentor. The biomass of dried cell was 2.5mg/ml, and the yield of carotenoids was 0.55ug/ml.
