Nonsteroidal antiinflammatory drug, probenecid, was linked with 2-hydroxyethyl methacrylate ( HEMA ) through covalent linkage. Initiated by azobisisobutyronitrile (AIBN), the drug-linked HEMA (abbreviated as HP) was homopolymerized and copolymerized with methy methyacrylate (MMA) to obtain polymeric drug in THE. HP polymeric nano-particles were prepared by emulsion copolymerization in water. To enhance polymer's hydrophilicity , amphiphilic graft copolymer containing probenecid was synthesized from radical copolymerization of HP with poly(ethylene oxide) (PEG) macromonomer. It was also synthesized from radical copolymerization of MMA with poly (ethylene oxide) macromonomer containing HP. Synthetic products were characterized with IR, 1H-NMR, GPC, DSC and TEM. Various experiment parameters controlling the copolymerization were studied in detail.
