The studies of novel method for the determination of polyphenols by high performance liquid chromatography with chemiluminescence detection were carried out in this dissertation, which consists of three parts.The first part is a review concerning the definition, the classification, the property and the analytical methods of polyphenol. Emphasis was placed on the development of high performance liquid chromatography determination of polyphenol, and then some research reports were given in the second part. Some kinds of polyphenols were separated and determined by high performance liquid chromatography with chemiluminescence detection, and therefore good results were obtained. The third part is a conclusion.1. Determination of Theanine in Tea Leaves by HPLC-Inhibited Chemiluminescencewith Co2+ CatalysisA method for the separation and determination of theanine in tea leaves was developed, based on inhibited chemiluminescence of cobalt-catalyzed luminol-H2O2 reaction by theanine. Separations were performed on an YWG-C18 column using a mobile phase of 0.01moI·L-1 acetic acid -sodium acetate buffer at pH 5.5 at a flow rate of 0.8 mL·min-1. The optimum concentrations of Co2+, Luminol, and fH2O2 were 2×10-9 g·mL-1, 2.5×10-4mol·L-1, and 5×l0-4mol·L-1, respectively. Under the conditions mentioned above, the relative peak area of inhibited chemiluminescence by theanine was linear with the concentration of theanine in the range of 0.0002 -0.005g·mL-1 and its regression equation was Y = 33862X+1.0605(r=0.9983). The average recovery was 97.82% with a RSD of 2.16%.2. Inhibited Chemiluminescence and HPLC Method for the Determination of Paracetamol in TabletsHigh performance liquid chromatography (HPLC) with chemiluminescence detection of paracetamol was developed based on its inhibitory chemiluminescence effect on a Luminol-ferricyanide system in alkaline solution. A detection limit (3) of 7.4x10-8 g/mL and a linear dynamic range of 8.0x10-7-1.6x10-5 g/mL can be obtained in the method A repetitive injection of 4.0x10-6 g/mL paracetamol solutions gave satisfactory reproducibility with a relative standard deviation of 3.4% (n=8). Contents of paracetamol in three locally available Pharmaceuticals were analyzed. From the obtained recovery of 98.6-101.4%, the feasibility of the developed method for real sample analysis was confirmed.3. Selective Determination of Tyrosine in Oral Liquid by High Performance Liquid Chromatography with Chemiluminescence detectionThe separation and determination of tyrosine in oral liquid by high performance liquid chromatography with KMnO4-Luminol chemiluminescence detection were investigated first time. The separation was carried out on Hypersil C18 column using the mobile phase of KH2PO4 (0.25mol/L)-CH3OH-H2O(l0:10:80,v/v/v). Under the optimum conditions of 3x10-5mol L-1 luminol.lx10-5mol L-1 KMnO4and 0.25 mol L-1 NaOH, the linear range and detection limit for tyrosine were l.25x10-6-5x10-5g/mL and 3.26x10-7g/mL, respectively. The relative standard derivation for 2.5x10-5g/mL tyrosine was 2.37%.4. Determination of Puerarin in Radix Puerariae by High Performance Liquid Chromatography with Chemiluminescence detectionThe separation and determination of puerarin in Radix Puerariae by high performance liquid chromatography with KMnO4-Luminol chemiluminescence detection were investigated first time. The separation was carried out on Hypersil C18 column using the mobile phase of KH2PO4(0.25mol/L) -CH3OH -H2O(10:15:75,v/v/v).Under the optimum conditions of 5x10-5mol L-1 luminol, 1x10-5mol L-1 KMnO4and 0.625 mol L-1 NaOH, the linear range and detection limit for puerarin were 0.0177-1.77ug/mL and 0.00367ug/mL, respectively. The relative standard derivation for 0.354ug/mL puerarin was 2.89%.5.
