| The study of interaction of small molecular compound and biologic big molecular has been the important research in the biochemistry. DNA is a kind of key biologic large molecule, it plays very important role in life genetic code' interpretation, transcription and copy. In order to discover and know DNA's character, structure, behavior and configuration, unveil life's secrete, people have been studied many kinds of metal complexes interaction with DNA. Especially for lanthanide ions and their complex and transition metale complexes interaction with DNA and RNA arose people's interesting. In this paper we studide the hydrolysis of oligodeoxyribonucleotide and pBR322 DNA by cerium ions and amino acid complexes, and then study the trinuclear Copper(â…¡) Complex cleavage the pBR322 DNA. Based on the hydrolysis of 26-mers oligodeoxyribonucleotide (ODN) by cerium ions, we studied the hydrolysis of oligodeoxyribonucleotide by cerium ions and amino acid complexes in physiological condition, according to the experiment, these amino acids can co-ordinate with cerium ion(â…£) to form complexes near the neutral condition (pH=7). At the same time, the denaturing ployacrylamidegel electrophoresis-silver dye method is used to analyze the hydrolysis of 26-mers oligodeoxyribonucleotide by cerium amino acid complexes round the neutral condition (pH=7). The results show that these cerium amino acid complexes can cleavage DNA at different degree, and this method is quick,safe and low charge. Based on this report we studied cleavage of pBR322 DNA by cerium nitrilotriacetate. The results indicated that this complex has an efficient cleavage for single strand of plasmid pBR322 DNA, making it from supercoiled form to open circular one.There is no report about the trinuclear Copper(â…¡) Complex interaction with DNA. We research the trinuclear copper(â…¡) complex [Cu3(L)2](ClO4)2 (where L = 1,3-(2-hydroxylbenzoylimino) propane) . The complex has been found to promote cleavage of pBR322 DNA from supercoiled form â… to the open circular form â…¡ and to linearized form â…¢, showing that the complex has higher cleaving efficiency to DNA. |
