Both Phosphatidyl Choline(PC) and Phosphatidyl Ethanolamine(PE) are important sorts of phospholipids, being widely used in the filed of food, pharmacy and so on. People pay more attention to the nature high-purity phospholipids because of its special property. At present, almost all of the high-purity phospholipids used in our country are imported,the annual demand is upward to several tons. So it is necessary to study how to produce high-purity phospholipids. The effects of water contents in stationary Phase and mobile phase on the simultaneous purification of PC and PE by column chromatography were studied systematically in this paper.At first, the process condition of extraction of phospholipids from egg yolk was investigated.The process included of lipid removal process with acetone,following with extraction process with ethanol,which provided good raw material of the following purification process. Then, the effects of water contents in silica-gel and mobile- phase on the simultaneous purification of PC and PE by silica-gel column chromatography were studied systematically in this thesis. The results showed that the water contents in silica-gel and mobile phase had great influence on the purification process. In the range of 10~12%(wt) for water content in silica-gel and 0.5~0.9%(vol) for water content in mobilephase, baseline resolution and higher recovery of PC and PE could be reached. The recovery for PC was more than 85% and PE over 70%.At the same time, the effects of water contents in aluminium oxide and mobile phase on the simultaneous purification of PC and PE by aluminium oxide column chromatography were also studied systematically in this thesis. We finded that the water contents in aluminium oxide has great influence on the purification process. In the range of 8%(wt) for water content in aluminium oxide and 0.1~l.l%(vol) for water content in mobile phase, baseline resolution and higher recovery of PC could be reached. The recovery for PC was more than 90% and PE over 40%.Not only could the work give some guides for the preparation of high purity PC and PE, but also it was very helpful in R&D of other natural products.
