Phospholipids are a kind of phosphorous-containing lipid,which mostly gets from soybean and egg yolk. Both Phosphatidyl Choline (PC) and Phosphatidyl Ethanolamine (PE) are important sorts of phospholipids, being widely used in the filed of food, pharmacy, cosmetic and so on. The nature high-purity phospholipids have become one of the hotspots for its special surface activity and emulsification.At present almost all of this production is imported in China, which makes the research on high-purity phospholipid preparation valuable.As a part of "Study on the Purification and Stability of Egg Lecition", in this paper, the breakthrough characteristic of PC and PE on silica gel column and aluminium oxide column was studied, in addition, the experiments about adsorption and desorption have partly done, all provide a basis for the establishment on separation technique of PC and PE. Moreover, the preliminary investigations have been made of the stability of lecition on different condition.The effect of mobile phase,sample concentration and flow rate on the process by silica gel column was investigated. From the results, PE as target products, the optimum operating condition was found that was 100%CHCl3 used as mobile phase, low sample concentration and flow rate were preferred. The sample concentration and the ratio of elution solution which influence the separation effect of PC and PE were further researched. According to the recovery and purity of PE and PC, 50mg/ml phospholipid solution and 10%MeOH as eluent was preferred. The purity of PE and PC was respectively more than 60% and 90%.The effect of mobile phase, sample concentration and flow rate on the process by aluminium oxide column was investigated. From the results, purifing of PC being the object, the optimum operating condition was found that 100%CH3OH as mobile phase, high sample concentration and flow rate were preferred. Based on the purity and efficiency, sample concentration (l00mg/ml) was confirmed. 100% MeOH was better to used to wash the column. In result, high-purity PC can be attained.Besides, the effect of medium, water contents of solvent, pH etc. on lecition stability was also studied. Experimental results showed that solvent can isolate the system from atmosphere, phospholipid solution was more stable than solid; pH played an important role in stability of phospholipid solution, here hydrosis was facilely occurred. It was indicated using of solvent won't acclerate the oxidation process, simultaneously water contents need be controlled.The work could give some guides in the preparation of high purity PC and PE. as well as separation and purification of other natural product.
