This thesis includes a review and a research section.In the review ,the principle, fabrication and characteristics of micro array chip and micro fluidic control chip are presented respectively. More over, the applications of the two kinds of chip are reviewed. Biochip which aims at analyzing the bio-analyte has become an increasing promising biological analysis method owing to its increasing sample throughput, improving performance and reducing the cost. We have also prospected that the ideal biochip system incorporating micro array and micro channels enables complete analysis, from sample introduction, to pretreatment, to the final analysis (separations, spectroscopy, etc.).The research section contains four subunits.In the first subsection, a novel sensor array chip for determination of hemoglobin in human blood has been developed based on chemiluminescence reaction of luminal and hydrogen peroxide catalyzed by hemoglobin and enhanced by p-iodophenal. The luminal and p-indophenols encapsulated sol-gel was spotted as an array of 25 spots at the surface of polymethyl methacrylate (PMMA) substrate. Each spot could be incubated in a 1μL of hydrogen peroxide and 1 μL of blood sample. The CL intensity of all spots measured simultaneously with CCD camera with 4 min. exposure. The present method has been applied for the determination of hemoglobin i,a blood and serum, and the results is compared with those obtained from standard method. The method has potential advantages of simplicity and high sensitivity with detection limit of 1.9ng, the relative standard deviation of less than 3%.In the second subsection, another sensor array chip for determination of benzoyl peroxide (BPO) in flour is described based on chemiluminescenct reaction of luminal and BPO catalyzed by Co2+. The two analytical reagents involved in the CL reaction, including cobalt ion and luminal, were immobilized electrostatically on cation-exchang resin and anion-exchang resin, respectively. BPO is determined by means of the reaction with cobalt ion and luminal, which were eluted by sodium sulphate. All 10 resins were put into the array fabricated in PMMA. The CL intensity of all spots measured simultaneously with CCD camera with 6 min. exposure. Thepresent method has been applied for the determination of BPO in flour. We also raise the theory basis of the course by means of kinetics experimentation. The linear range of the benzoyl peroxide concentration was 1×10-4 g/mL-l×10-6 g/mL. The detection limit was l×10-6g/mL and the R.S.D for 7 spots is 5.6%.In the third subsection, a rapid and sensitive chemiluminescence (CL) microfluidic chip fabricated in PMMA for determination of benzoyl peroxide in flour is described. The width and the depth of the microchannel were all 300um in the chip(50×40×5mm). Benzoyl peroxide can directly oxidize luminal to produce chemiluminescence. Two sampling structures of double-tee sampling structure and microvalve sampling structure were used. The sampling volume was 0.72uL for the double-tee sampling structure and 0.79μL for the microvalve sampling structure. The precision of the two sampling structures was almost the same. The linear range of the benzoyl peroxide concentration was 8×10-7-1×10-4g/mL. The detection limit was 4×10-7g/mL and the R.S.D for 8 times is 3.06%. The proposed method has been successfully applied to the determination of benzoyl peroxide in flour.In the last subsection, a chemiluminescence (CL) microfluidic chip fabricated in PMMA for determination of hypochlorite in tap water is described in this paper. The microchip fabricated in PMMA was readily produced in our laboratory. The width and the depth of the microchannel were all 300 (am. Luminal was moved through the channels using a injection pump and the sampling volume was 0.72μL every time. Hypochlorite can directly oxidize luminal to produce chemiluminescence.
