Microfluidic chip (microchip) has attracted increasing attention because it can beintegrated with separation channel, microvalve, micropump and detector etc, and canperform almost all operations in ordinary laboratory. However, separation still is oneof the most important functions for microchip. Therefore, how to improve theefficiency of seperation and fabrication of microfluidic chip is still one of main goalsfor study. The main points of this thesis including:The first chapter is a brief overview on the progress in the microfluidic chipstudy including fabrication technique, separation modes, detection methods and itsapplication.In the second chapter, a new technique of fabricating glass microfluidic chip within-dium tin oxide (ITO) glass was described. The (ITO) glass microfluidic chipshowed excellent surface properties, such as stable EOF and good light transmission.The technique of ITO microfluidic chip fabrication simplified the fabrication process,shortens the fabrication time and decreases the costs of microchip.In the third chapter, a method for the separation and detection of fivecasomorphin peptides by microfluidic chip together with laser induced fluorescence(MEC-LIF) was described. Using fluorescein isothiocyanate (FITC) as thepre-column derivatization reagent, a MEC-LIF method was established for thedetection of five casomorphin peptides. The derivatization and separation conditionshad been optimized. Finally, the methods were applied to the analysis of casomorphinpeptides in cheese which was decomposed with the protease enzyme.In the fourth chapter, a monolithic columns microfluidic chip formed withsol-gel process and polymethacrylate polymerization was described. The monolithiccolumn microchips were used to separate isomers, namely catechol and hydroquinone.A new ‘three-T’ sample injection and a convenient interface device were designed tomatch the monolithic column microfluidic chip. The device was used to separate5-HTP, DA and5-HT in the chip with polymethacrylate monolithic column.
