Polygonum hydropoper L. is wide range in China and has wide use in medicine, edible, and natural pesticide etc. In order to comprehensive utilize the precious plant resource, we studied the extraction, separation, and identification of chemical composition in Polygonum hydropoper L. in this paper.The chemical component of a plant relate to the region and climate, so we detected the chemical constituent of Polygonum hydropoper L. from JiangXi Province by system pre-examination. The detection result demonstrated that the Polygonum hydropoper L. contain protein, organic acid, carbohydrates, tannins, flavones, volatile oil, but no cardiac glycoside, coumarin, alkaloid and saponin steroids. The contents of the principal chemical component was also detected; there was 3.13% tannins, 2.63% flavones, 0.32% volatile oil in the herb, and 0.94% volatile oil in the leave of Polygonum hydropoper L.The standard curve of flavones was plotted by using spectrophotometry, and the regression equation y = 0.0247+7.9456.x was obtained which correlation coefficient was r =0.9999. The study about the extraction technology of flavones in Polygonum hydropoper L. indicated that the flavones can be obtained 2.98% by the optimum technology. Five monomers were separated by polyamide column chromatography, they are Rutin, Hyperin, Quercetin, Isorhamnetin and Kaempferol. Isorhamnetin was firstly separated from Polygonum hydropoper L.The dissertation studied firstly the separation of Polygonum hydropoper L. volatile oil with supercritical carbon dioxide extraction. The optimum technique condition was determined as follows: (1) the pressure was 25MPa; (2) the temperature was 40℃; (3) the time was 120min. Under such conditions above, the final yield of volatile oil could be maintained 3.72%. The main chemical constituent of the Polygonum hydropoper L. leaf volatile oil which composed the whole volatile oil's 80.66% was analysed by GC-MS firstly, and the 8 chemical components have never be reported in Polygonum hydropoper L previously.The chemical compositions and the method of extraction and separation was studied systematically, The result of this research will offer reliable information for Polygonum hydropoper's application.
