To improve the biotransformation technology, and upgrade Non-aqueous Enzymology research, and improve the quality of Ethyl Hexanoate, the immobilization of Lipase, the synthesis of Ethyl Hexanoate by immobilized Lipase and Ethyl Hexanoate extraction were studied.Main points are as follows:1. The comparison of ester synthesis by microbial Lipase in different media (water, solvent, solvent- free solution) showed that the results in synthesis were better in solvent phase than in water and solvent -free solution. It was identified that the Candida sp Lipase is best in the reaction of biocatalysts Ethyl Hexanoate four kinds of Lipase. And choosing proper Lipase sources was the reaction's key.2. The Immobilization of Lipase: In the organic solvent immobilization of lipase was studied .The immobilization conditions such as supporting materials, water content, organic solvent, the time of immobilization and the amount of enzyme added were determined, The activity recovery of immobilized lipase is 62%, The immobilized lipase is stability and its operational half-life is about 240 hours.3. The synthesis of Ethyl Hexanoate by Immobilized Lipase: The effects of facts on esterification of Hexanoic acid and ethanol by immobilized lipase in solvent have been studied, such as water content, solvents, reaction temperature, amounts of lipase added et al. After 8 h, The conversion yield in Ethyl Hexanoate synthesis by immobilized lipase was 86.69% in the Cyclihexane with 0.1g lipase, and 0.4% of water, added 1.0g of molecular sieve at 30 C.4. Ethyl Hexanoate extraction: On basis of comparing product-collecting methods, vacuum distilling was selected and used successfully. By IR and GC, the resulting sample was detected and identified as Ethyl Hexanoate. The analysis of product shows that the quality items are in keeping with the National Standard for Food Additive Ethyl Hexanoate. Compared with Ethyl Hexanoate from chemical synthesis, the heavy metal in that from enzymatic esterification, such as As and Pd, are in very low concentration.
