Studies On Isolation, Purification And Activity Of Two Compositions: Polysaccharides From Chinensis Actinidia Planch And Water Extracts From Onion

An anti-tumor polysaccharide was isolated and purified from Chinensis Actinidia Planch. The soluble fraction of onion was extracted and the activity of lowering cholesterol, was tested.One raw polysaccharide was isolated from the juice of the fruit, by trichloraceticacid (TCA) precipitation, ethanol precipitation, decolor and vacuum freeze-drying. Using ion exchange chromatography in packed Zhengguang DEAE-agarose column, the raw polysaccharide was separated into two parts, one was an acidic polysaccharide from the elution process, and the other, a neutral polysaccharide, was a non-bound substance. Another raw polysaccharide was isolated from the fruit, by hot-water extraction, ethanol precipitation, cetyltrimethylammonium bromide (CTAB) precipitation and vacuum-drying. Using ion exchange chromatography in packed DEAE-Sepharose FF column, the raw polysaccharide was separated into two parts in elution process, in comparison of the gradient of salt concentration and the conductivity curve with the conductivity detector of chromatography system. The results of size exclusion chromatography showed that the molecular weights of the two parts were similar.The inhibitory effects of the raw, the acidic and the neutral polysaccharides on the transplanted tumor cells of Sarcinoma 180 (S180) in mice were tested. The results showed that with the dosage of 150mg/kg the acidic polysaccharide could evidently reduce the tumor weight of S180, and the inhibitory rate reached 54.2%. But the others could not inhibit the S180.The soluble fraction of onion was isolated by water extract and the activity of lowing cholesterol on the hyperlipidemic mice was tested. The results showed that the water extract could decrease the level of blood cholesterol. After ethanol precipitation or adding some powder of ginseng, the effect was not observed.
