After the wheat germ was degreased by SFECO2, crude Polysaccharides was extracted with water, acidic liquid and alkali liquid respectively from defatted wheat germ. Crude Polysaccharides was precipitated by alcohol with different density and purified with different methods of deproteinization. As a result, the best extracting method and purifying method have been chosen and the extracting technology has been optimized. The studies have shown that the water extraction was the best method. The parameters of extracting technology were as follows.The ratio of solid to water was 1:8;the extractive temperature was 90℃; extractive time was 2.5 h; extractive times was 3 times. The precipitation by 85% alcohol was economical. Precipitating the protein in the wheat germ by adjusting pH to 4.5 was the best method.Through classifing the purifying Polysaccharides by Sephadex-G100, two types of Polysaccharides have been achieved. After acidic hydrolysis, and assayed by HPLC, the wheat germ PolysaccharideⅡ was mainly consisted of Glucose,xylose and arabinose. The further results of studies have shown that the wheat germ Polysaccharide could promote the growth of Bifidobacterium , obviously inhibit the lipoperoxidation and enhance the endurance of rat to adversity.
