Ginkgo biloba is an ancient tree which, for centuries, the Chinese have cultivated and the leaves of its have been as a Chinese traditional medicine for hundreds years. Ginkgo biloba tree date back than 200 million years and it has been entitled "living fossil". In recent decades, concentrated extract of Ginkgo biloba leaves have been marketed in Western countries as herbal medicines which have abroad bioactivity such as enhancing cerebral blood flow, improving memory, and as a neuroprotectant. Ginkgo biloba leaves contain a lot of flavonoids and ginkgolides. But their bioavailability is not high for their molecules structure limited. According to many literatures report, under certain condition natural active component integrate with phospholipid into phytosome or phospholipid-complex, which physical and chemistry properties change greatly, more than that its lipophile is great, it means that it can improve the absorption in vivo, apparently change its biology effectiveness.This study extracts rutin from flower of Japanese pagoda tree, and hydrolyzed it to obtain quercetin. Investigate related factor such as pH, time in the procedure. Determined the content of quercetin by HPLC and identify with standard in IR, TLC, characteristic reaction. The hydrolyzed condition is 2.0% HaSCM, refluxing for 30min.And manufacture the phytosome with quercetin as one of the represent of Ginkgo flavones. Certainly made corresponded comparison.Investigate the physical and chemical properties of quercetin, lecithin, physical mixture and phytosome with FT-IR, XRD, DSC, and SEM. There are distinct differences between phytosome and physical mixture. Compared with quercetin and lecithin, through these facts from above examinations, phytosome has some new clear physical and chemical properties. It may be result from the coalescent effect between quercetin and polarity group of lecithin. It is a new drug amorphous phase and/or inclusion complex.Investigated the apparent oil/water partition coefficient (P) of free quercetin and quercetin in phospholipid complex in different pH waters and n-octyl alcohol. The result shows that: the dissolve capacities of quercetin compounded with lecithin heighten nearly 20times than free quercetin and the coefficient (P) rise 10 times. It isbecause lecithin can improve the dissolve of quercetin in n-octyl alcohol. The dissolve curve show that pattern of quercetin was as same as its phospholipid-complex. Compare the stability of phospholipid complex with that of quercetin in certain temperature and damp condition. The results show that we should keep it away moist and hot.Through all above research: ginkgo flavones interface with phospholipid. uniformity dispersion in phospholipid and show new physical and chemical properties, such as the change of peak value of IR, the disappearance of 20 of quercetin crystal, and the variety of DSC peak and the rising of apparent oil/water partition coefficient (P). Combined with pharmacology examination results, the ginkgo phytosome can improve the activity and availability of extract of ginkgo biloba.In present study, with analytical methodology research the content of flavones in phospholipid-complex was determined by HPLC and identified the ginkgolides A, B and Bilobalide with TLC then formulated the quality criterion of Ginkgo phytosome. On the basis founded the formula of complex rate of ginkgo phytosome. With the guidance of compound rate to optimize the prepared condition of Ginkgo-phospholipid complex. The study was carried out through orthogonal test. The result shows ratio 1 :2,reaction 20h in THF solvent.
