Gracilaria tenuispitata var. liui were collected from the cultivate base of algae in Zhanjiang city, Guangdong province. The processing conditions of extraction of Gracilaria tenuispitata var. 1 iui polysaccharides ( GTSP) were optimized. We also studied the antiviral activities against HSV-1 of the above SP samples.The effects of processing conditions on the extraction of GTSP were respectively studied as guiding by the extraction rate of GTSP . By L9(34) orthogonal experiment design, the optimum processing conditions of extraction were determined. They were as follows: temperature of extracting 80 , volume of water 70 folds, time of extracting 3 hours .The sulfated polysaccharides(SP) were extracted with hot water and sodium hydroxide solution from Gracilaria tenuispitata var. liui . They were separated by using DEAE-cellulose column chromatography. Their structures were determined by IR and GC-MS. The contents of the total polysaccharides, 3,6-anhydrogalactose and sulfate were analyzed by the methods of anthranone-sulfuric acid, resorcin and BaCl2 turbidimetry.The antiviral effects of all samples on HSV-1 and their cytotoxicity to Vero cell were studied by CPE and MTT method. The results showed all of samples can not kill the above viruses directly. The cytotoxicity of samples on Vero cell was not detected.. Compared with other algaesulfated polysaccharides, it can be inferred that the antiviral activity of sample is correlated with the kind of polysaccharide and the content of sulfated groups.The extraction of cilaria tenuispitata var.liui from ethanol was separated as guiding by the biological activities. Two sterols,cholesterol(D,cholesta-4,6-dien-3 p" -ol(2) were identified by GC-MS. MIC of the compound to Escherichia coli is 1.25mg/mL.
