| In recent years,the populations suffering food allergies have continued to rise,which has seriously endangered daily lives of patients.And the issue has caused worldwide concern.In order to control the continuous increase of food-allergic populations,it has becomed a hotspot to develop a safer kind of health-care products in related fields.Many studies showed that many kinds of marine algae have varieties of biological activities,among which Gracilaria lemaneiformis sulfated polysaccharides and Porphyran sulfated polysaccharides have immunomodulatory effects preventing food-allergic diseases.On the other hand,effervescent tablets can disintegrate rapidly with the characteristics of high bioavailability and to be carried easily.Thus,it will be an innovative choice for the application of effervescent tablets upon the antiallergic fields.In this paper,with Gracilaria lemaneiformis sulfated polysaccharides and Porphyran sulfated polysaccharides,prepare a kind of red algae sulfated polysaccharides effervescent tablets with antiallergic activity,which would be studied subsequently.In this study,Gracilaria lemaneiformis sulfated polysaccharides and Porphyran sulfated polysaccharides were extracted by high temperature and pressure,and alcohol precipitation.The contents of total carbohydrate were 97.04% and 98.17% respectively.The proportions of deoxidize carbohydrate were 31.52% and 25.18% respectively.The viscosities were 0.08 dL/g and 0.10 dL/g respectively.The sulfate contents were 10.16% and 12.34% respectively.The two red algae sulfated polysaccharides both showed sulfate active groups by the Fourier transform infrared spectroscopy.The red algae sulfated polysaccharides effervescent tablets were prepared by acid-alkali granulation separately.The ultimate formula(mass percent)of effervescent tablets was as follows: Gracilaria lemaneiformis sulfated polysaccharides 5%,Porphyran sulfated polysaccharides 5%,lactose 30%,mannitol 13%,citric acid 10%,tartaric acid 10%,sodium bicarbonate 20%,stevioside 3%,PVP-K30 4%.The formulation has a particle size of 26.40% and an angle of repose of 23.62 °.The formed tablet has a weight of 0.54 g and a disintegration time of 53.33 s.The cell model in vitro and murine model in vivo were established to study the anti-food allergic activity of the red algae sulfated polysaccharides effervescent tablets.The results of cell experiments in vitro showed that the red algae sulfated polysaccharides effervescent tablets could inhibit the degranulation and histamine release of RBL-2H3 cells.The results of mice experiments showed that the red algae sulfated polysaccharides effervescent tablets can alleviate the allergic symptoms of mice,inhibit the levels of of OVA-specific IgE,histamine and mast cell protease-1,inhibit the levels of Th2 cytokines,promote the levels of Th1 cytokines and Treg-associated IL-10 and TGF-β.The results of flow cytometry showed that the red algae sulfated polysaccharides effervescent tablets can inhibit the numbers of DC cells,Th cells,B cells and MC cells,and the red algae sulfated polysaccharides effervescent tablets can promote the numbers of Treg cells additionally.In summary,the red algae sulfate polysaccharides effervescent tablets were successfully prepared,and the anti-food allergic activity and its mechanisms of the red algae sulfate polysaccharides effervescent tablets were studied,which could provide references and supports for the development of related tablets. |
