Tyrosinase is a major enzyme, which controls the speed of melanin biosynthesis. Usingpharmaceutical chemical materials to penetrate the hair cell, the generating of hair melanincan be promoted. Thus the goal of darkling hair can be obtained. Although several materials,which can activate or prohibit the tyrosinase have been researched, especially some materialswhich can prohibit the tyrosinase were founded. The research of materials using in cosmeticsfield, which can activate tyrosinase, is still limited.The paper is examined five Chinese herbs, Salvia miltorirrhiza Bge, Mentha haplocalyxBriq, Scutellaria baicalensis Georgi, Taraxacum mongolicum Hand-mazz, Prunella vulgaris L.After the process of separation, extraction, drying, the concentrate materials can be obtained.The activating ratio of tyrosinase is tested by absorption spectrometer. The results show thatthe extract material of Salvia miltorirrhiza Bge and Mentha haplocalyx Briq have betteractivating ratios of tyrosinase than others.The crude concentrate materials can be gotten through the process of extraction,concentration from the metha's water extracts and Salvia miltorirrhiza's activating ratio oftyrosinase is tested toughly. The result shows that the butyl alcohol extracts of metha's watersolution has the good activating ratio of tyrosinase and the ethyl ether extracts of Salviamiltorirrhiza's methanol concentrates has the good performance of activating tyrosinase,through the forward silica gel column layer chromatography elution analysis of butyl alcoholextracts of mentha's water solution and ethyl ether extracts of Salvia miltorirrhiza's methanolconcentrates, more than 10 eluants can be obtained. To examine these eluants' activating ratioof tyrosinase, the results proved that the eluants from mentha's water solution is not satisfied,because its activating ratio of tyrosinase is only 19.11%. On the contrary, the eluants fromSalvia miltorirrhiza'smethnol extracts, especially No.3a has a remarkable activating ratio oftyrosinase, up to 266%, which is far better than metha's water solution.Thus we use Salvia miltorirrhiza as raw materials. Extraction process was carried out byseveral different solvents. Each extraction materials to the activating ratio of tyrosinase wereexamined carefully. It proved that the ethyl ether extracts had better activating ratio oftyrosinase, but lower extracting ratio. At the same time, using methanol as extracting agent, itsethyl ether extracts had not only good activating ratio of tyrosinase, but also high extractingratio, which is better than Salvia miltorirrhiza's ethyl ether extracts.Drying ethyl ether extracts in low temperature, firstly building it with Cu2+ ions,cysteine and panaxcoside separately, measuring their activating ratio of tyrosinase, the rationshowed 396%, 170.93% and 143.10%, which was far better than pure ethyl ether extracts,then, building ethyl ether extracts with Cu2+ ions, cysteine and panaxcoside together indifferent concentration and measuring their activating ratio of tyrosinase, the results alsoproved better than pure ethyl ether extracts, activating ratio of tyrosinase.To examining the efficient block of activating ratio of tyrosinase, the TLC process wascarried out for methanol extract No.3a. The result showed its had not adsorption in 365nm,but only had weak adsorption in 280nm. Acting 280nm as basic wavelength, to explore theseparation condition of HPLC, examine its detail component part;we found methanol extractNo.3a could be separated by HPLC.The methanol extract No.3a was purified and separated by HPLC, five pure separatingagents were obtained. To measuring these agents, we found No.5 had the best activating ratioof tyrosinase. Through the process of IR, UV, 1HNMR, 13CNMR and MS, its constructer wasidentified as cryptotanshinone.
