According to the research result before, the plant polysaccharides did benefit for food intake on the diabetes suffer .The polysaccharides of of elderberry and ophiopogon japonicus ker-grant is very effective significantly reduce blood glucose in the diabetes rats.maybe it is the same to human kind.The extraction rate of traditional method of hot water extracting and ethanol precipitating was always very low though the plant polysacchride possess wonderful health function.this study might reveal how to extract water-soluble polysaccharide from Elderberry by different methods.and several important factors to extraction were discussed.Water temperature(80,90,100℃),volume(40/l, 50/1, 60/1) and extracting time(3,4,5hour) have been selected to be three key factors in Elderberry water-extracting direct cross experimentwe have gotten the optimum technics parameters in the water-extracting after analysed the yield of Elderberry polysaccharide.So the same materials were adding to acidity water and alkalescence water followed the optimum technics parameters.and we also treating Elderberry with ultrasonic and using a united technology of the combination of cellulose-enzyme decomposing and water -extracting methods to obtain the Elderberry polysaccharide.The result is:A method suitanble for just hot-water extracting Elderberry was adding 60 timers of water volume as much as that of sample extracting 3 hours at 100℃,but the optimum conditions of extracting SWH were as follow: adding 60 timers of water volume as much as that of sample .extracting 3 hours at 100XH and adding cellulose-enzyme 1.5% weight of target matter concentration, extracting 180 minutes at 40℃(pH5.0) and residue extracting was the same as the first condition, this method could enhance the efficiency of extraction to 15.36%,which is 37.8% higher than the traditional methods.Then we found that there are abundant protein in the course of Elderberry purification.so we campared with four normal removing- protein methods such as most familiar sevage method ^ TCA method—be served the turn to plant polysaccharide, papain method ,we are concern with the efficiency of the removing method and the yield of polysaccharidcAnd the result was just follow as: the optimum conditions of removing protein were as follow: adding papain 0.1% weight of target matter concerntration .removing 120 minutes at 50℃( p H4.0) and using TCA method(tca concerntration 2%,TCA solution/butanol/polysaccharide solution=l/3/5volume) to remove protein.
