Polysaccharide is a very important substance of bioactivity in living creature or plants. Because of owing a variety of bioactivities and extensive usages on functional food and medicine, the study on polysaccharide attracts more and more attention. Pumpkin polysaccharide is a kind of polysaccharide of bioactivity, however, the study on pumpkin polysaccharide are few. So we study the extraction and purification of pumpkin polysaccharide and its characters and hope to find the theories to improve its applications. The main experiments were as follows:1. Pumpkin polysaccharide was extracted from pumpkin by means of hot water extraction. The factors that affected the extraction of pumpkin polysaccharide included the ratio of material to solvent, extracting time, temperature and times of extracting. A four-factor three-level orthogonal experiment was designed according to the single factor experiments. The multi-linear equation showed that the optimal conditions were at the temperature of 80℃, extracting time 3.2h, the ratio of material to solvent 1:40 and times of extracting 3. The biggest influence factor was temperature, and then extracting time and the ratio of material to solvent, the smallest influence factor was times of extracting.2. The experiment of purifying pumpkin polysaccharide was studied. As to the choice of precipitant, ethanol was better than acetone. When using ethanol to precipitate pumpkin polysaccharide, the ratio of concentrated solution to ethanol was 1:4. Activated charcoal was used as decolourant; the influence sequence was the activated charcoal's consumption, temperature and time. The best conditions of decolonization were under the temperature of 20℃,consumption of activated charcoal 0.83%,time 5 minutes. The process of deprotein was carried out after adding papain. It was found that the goal of deprotein was achieved by shaking method with solution, ratio of chloroform to butanol 4:1. Pumpkin polysaccharide was composed of glucose, isodulcitol, arabinose and galactose. 58.2% pumpkin polysaccharide could be get.3.
