Due to its rich biological function and active, polysaccharide has been widely attention.Balsam pear and pumpkin as a daily diet of vegetables, not only delicious and nutrition, they also have the effect of anti-tumor and antioxidant, etc. Because of their good effect, they have the very important research value and development potential. However, the polysaccharide extracted directly separated from balsam pear and pumpkin,can not meet the requirements of the medical treatment,it makes that modified polysaccharide structure has become very necessary, sulfated modification as a simple and effective method is currently subject to people’s attention.This paper evaluated the physical and chemical properties of the W-MCP, S-MCP, WPP and their sulfated modification polysaccharide, using infrared absorption spectroscopy and atomic force microscope measure their structure change, compare the change of antitumor and antioxidant activity, evaluation the affect of sulfated modification on polysaccharide structure and biological activity.Through research, obtains the following conclusions:(1)The tests of physical and chemical properties of W-MCP, S-MCP, WPP show that:sulfated modification makes W-MCP from pale yellow loose flocculent structure into gray flocculent, makes S-MCP from white fluffy gray powder, makes WPP from light yellow flocculent wool into beige powder.Barium sulfate turbidimetry measure the degree of substitution of SW-MCP, SS-MCP and S-WPP are0.93,0.62,1.02, respectively. The chemical tests show that SW-MCP, SS-MCP, and S-WPP containing sulfate groups, SO42-combine with the hydroxy of polysaccharide and generate lipid key, sulfated modification is successful.(2)Infrared spectrum scanning on six kinds of polysaccharide, the results show that the infrared spectrum of the three kinds of sulfate modified polysaccharide emerge the characteristic absorption peak of the S=O and C-O-S key, or the characteristic absorption peak become strong, described SW-MCP, SS-MCP and S-WPP have been sulfated.(3)Atomic force microscope imaging on six kinds of polysaccharide, the result show that:W-MCP contains cyclic stereo structure, aggregation degree is low; S-MCP mainly exist in branched chain structure, there is interaction between branched chain. WPP contains pyran ring, structure aggregation degree is high. Sulfated modification makea three polysaccharides become more decentralized in space, loose, and makes molecular structure smaller. WPP’s the most obvious change. The experiment shows that the sulfated modification changed the spatial structure of polysaccharide, makes the structure of the polysaccharide more spread out, smaller molecules.(4)The inhibition activity of six kinds of polysaccharide on the proliferation of human leukemia cell line K562was measured by using MTT assay.The results showe that the six kinds of polysaccharide has certain inhibitory activity on K562, maximum inhibition rate is:W-MCP was28.07%, S-MCP was25.42%, WPP was33.26%, SW-MCP was37.14%, the SS-MCP was29.88%, the S-WPP is40.13%, respectively,and S-WPP has the highest activity. The experimental results show that both balsam pear polysaccharide and pumpkin polysaccharide have anti-tumour activity,and sulfated modification can improve the anti-tumor activity of polysaccharides.(5)Using four experiments (clearing O2-·, OH·, DPPH-, inhibition of lipid peroxidation) inspects the antioxidant capacity of six kinds of polysaccharide. The results show that:the sulfated modification can effectively improve the ability of SW-MCP and SS-MCP clearing O2-·,and can make the S-WPP have the new ability to clear O2-·. Sulfated modification can effectively improve the ability of clearing OH· of the W-MCP and S-MCP, but make the ability of WPP clearing OH· weaker; Sulfated modification can significantly improve the ability of SS-MCP clearing DPPH·, can increase the ability of SW-MCP clearing DPPH·at high concentrations, while the ability of S-WPP clearing DPPH· do not help.Experiments show that with the difference of the kinds of polysaccharide and free radicals, the antioxidant activity of sulfated modification changes at different levels.
