In this thesis, some derivatives of aniline such as fuchsin basic (FB), o-aminophenol (OAP); o-dianisidine(ODA) were selected as the studying objects. The study was involved in spectroscopy, electrochemistry and the interaction of FB with DNA; the fabrication, the electrocatalysis of poly(o-aminophenol)/nickel modified platinum electrode(Ni/P-OAP/Pt)) and the immobilization of DNA on Ni/P-OAP/Pt; and the thin-layer spectroeletrochemistry of ODA at a platinum minigrid optically transparent electrode. The interactions of DNA with fuchsin basic (FB) were studied by spectroscopic and electrochemical methods. The more powerful interaction of FB with dsDNA than with ssDNA was showed in the experiments, which may be expected to be applied in recognition of dsDNA with ssDNA and in DNA biosensor as hybridization indicator. (Poly) fuchsin basic modified electrode was prepared and the electrochemical behavior of DNA on such an electrode was also studied.Poly(o-aminophenol)/nickel modified platinum electrode(Ni/P-OAP/Pt) was prepared by cyclic voltammetric electropolymerization in HClO4 solution. DsDNA was immobilized onto such electrode via three-dimensional structure of polymer film and electrostatic effect between dsDNA and Ni2+ ion. At the same time, the Ni/P-OAP/Pt electrode exhibited an electro catalytic effect on the glucose oxidation. The electrocatalytic oxidation peak current of glucose has a good linear dependence on concentration of glucose.
