A Study On The Technology Of Synthesis Of Chiral Phenylglycinol

Chiral aminoalcohol is an important kind of chiral substance in the asymmetric synthesis, meanwhile, it is widely used in the field of medicine synthesis and the resolution of raceme. In view of those properties, chiral aminoalcohol's serial products are steadily on the increase and show the great capacity of the potential market in the world. Chiral phenylglycinol has more wide application value in the utility of chiral aminoalcohols.The technology of the synthesis of chiral phenylglycinol is an important emphasis on my thesis. Having consulted a lot of associated documents at home and abroad, we have designed a new synthesis method of chiral phenylglycinol which can directly make the chiral phenylglycine into chiral phenylglycinol through the compounding reducing agent of good properties, and the kind of compounding reducing agent which is made from (A) metal hydride and (B) Lewis acid has been worked out through the theoretical analysis and experimental selection, At last, we gain a high yield and high optical active purity of phenylglycinol through the small scale test and pilot scale experiment. Then, we have succeeded in conversion of experiment scale. Consequently, we have laid the foundation for the industrialization of phenylglycinol. The optical purity and structure of product—chiral phenylglycinol have been confirmed by the HPLC, the polarimeter, and the infreded spectrum of graph.The technology of preparation of chiral phenylglycinol from chiral phenylglycine is turned out to be a technical platform which can prepare many kinds of aminoalcohol through repeated experiments of the small scale test.On the basis of above work, we also have designed the HPLC method of purity determination. The method has never been seen on the associated documents. Thus, our work has been well ready for the draft of the enterprise standard of chiral phenylglycinol.
