| Depression is one of spirit obstacle disease accompanied by sentiment. Its symptom shows gloomy, taciturnity, unresponsive and even attempt to suicide. The number of people who suffer depression are increasing since the survival press are added and emergency are pricked up while the modern life rhythm are hurried up. Now depression is the fourth grave disease among the 10 topic serious diseases in the world. It is estimated that depression will become the second serious disease in 2020.There are also found serious side and toxicity effect when we using chemical drug in depression clinical therapy, minimum the chemical drug application scope and foreground. Our studies are based on finding effective compound from the fruits of Tribulus terrestris. We have found that the total saponins of the fruits of Tribulus terrestris are the antidepression substance. Then we developed the extraction and purification technology of the total saponins. Further more, we develop the preparation technology of the effective compound to produce a kind Chinese medicine tablet, which its disintergrants and drug release ability are similar to chemical tablet.Firstly we establish an animal model of depression. The model was used to screen the antidepression effective compounds of the the fruits of Tribulus terrestris while we found the total saponins are the antidepression substances. To further study its antidepression mechanism of th total saponins, we measured the nerves agent such NE, 5 —HT, 5 —HIAA and DA in the brains of the animal that administered physiological saline, total saponins and Sertraline Hydrochloride respectively. Results showed that the total saponins were the antidepression substance work on several ways, and the high dose of total saponins (200mg/kg/d) can increase the 5-HT content of the animal brain, similar to the controlling drug-Sertraline Hydrochloride . This result also indicates that the antidepression approach between the high dose and low dose total saponins may be different.After the total saponins determined as the antidepression substance, we proceed to optimized the technology for extracting that total saponins from the fruits of Tribulus terrestris. Based on the test of the influence factors, we use uniform design to screen the parameters of circumfluent extration. The optimal parameters of the circumfluent extration were that soaked in 75%EtOH(8 folds) for 40 minutes, then circumfluent extracted for 2.5h, 3 times. In the purifying technology, we selected macroporous adsorption resin to purify the total saponins. The process study showed that HPDIOO macroporous adsorption resin had the highest adsorption ratio among four resins(HPD100> D10K HPD100a> HPD300). The experiments showed that the optimal parameters of purification were original concention as 10—15mg/ml,flow velocity as 2BV/h,and 6BV water, 2BV 30% EtOH, 4BV 70% EtOH as the eluting solvent respectively, gathering the part of 70% EtOH. The purity of total saponins we get are near to 60%,and the refine degree could reach 271.94 %.Based on the extracted and purified technology of the total saponins, we produced extracting material, and established the quality control of extracting material. We set out the extracting material's Micromeritics character and solubility. At last, we found the extracting material possess good quality and stability through stability test.In the preparation process, we refer the tablet criterion of chemical drug as the reference criterion of tablet we would to prepare to produce. We selected the best formulation and technics by the orthogonal design and single factor experiment,. The tablet we produce can be disintegrated within 10 minutes and drug released more than 70% within 20 minutes. Its drug releasing mechanism included matrix eroding process and diffusing process. The tablets showed stable content of effective composition and good stability.
