A strain producing chitosanase was isolated from soil samples and named K-9. The result of the 16srDNA sequence alignment is that it had 99% homologic with Ralstonia basilensis The optimal fermentation conditions for K-9.producing chitosanase were incubated at 30℃, in 500ml Erlenmeyer flask containing 100ml medium (pH 6.0) with 1.0% chitosan as substrate. The chitosanase activity of fermented broth was 1.36U/ml when K-19 was cultured under the optimal conditionsat 4d. A inducible and extracellular chitosanases secreted by K-9 were purified by ammonium sulfate precipitation as well as gel filtration. The purified enzymes had molecular masses of 41.3kDa by SDS-PAGE. The chitosanase have an acidic pH optimum, which were 4.0. The enzymes were stable over the pH range 3 to 5. The optimum temperatures for hydrolysis of the chitosanases were determined to be 40 ℃, Metal ions of Mn2+, Na+ K+ accelerate the enzymes activity and Cu2+, Fe3+and Ag+ inhibited activity,...
