The maize is an important grain crop, and used as fodder in the world from 1998, the gross output of the maize has already exceeded that of the wheat and rice and become the first largest crop in the world. Generally, the essential amino acid content of the normal maize (lysine, albumen, tryptophan) is very low, and the lysine is the first restricted amino acid in the nutritive value of the maize. It is an important goal that improved the protein quality and increased the lysine content for the maize breeding.Using large sample DNA of capacity technology, the maize high lysine content of SSR mark analyse technological platform has been established in this paper. The main results are as follows: the five comparing research methods of drawing maize DNA mark have shown that the method of iso- sulphur cyanic acid guanidine drawing maize genome DNA has the superiority of using less medicine, operating sequence more simple , faster, and costs lower , steady and reliable quality of DNA, etc..Utilizing this method , one person can draw 300-400 DNA samples each workday.lt indicates that the method can be used in SSR marker of maize,.Utilizing SSR marker phi057, phi 112 and umc1066, the polymorphism between normal maize inbreds and o2 maize inbreds has been analyzed. The results show: SSR marker phi 112 is dominant marker between normal maize inbreds and QPM inbreds. A band of 420 bp in DNA PCR amplified product of QPM inbreds is absent, which is obviously deferent from normal maize inbrede . phi057 and umc1066 are co-dominant marker between normal maize inbreds and QPM inbreds .Taking phi057 as the inducer, 430 bp of DNA band can be amplified in normal maize inbreds,and 440kb in QPM inbreds;Taking umc1066 as the inducer,the results are 460 bp and 480 bp.Using phio57 marker to test BC1F1,the separate regulation of O2O2 genotype and o2o2 genotype accord with Meng Del heredity separate proportion (1:1 ). Studying two genotype of maize lysine content and seed of grain, we can find that O2O2 maize seed grains of lysine content (0. 428%) is notably higher than that of O2o2 (0. 333%). It indicates that using SSR phio57 marker to help selecting o2o2 genotype can improve the efficiency of selecting PQM maize.
