Selenium is an essential trace element for human body. It can be used as anti-cancer, cancer prevention, heart protection, prevention and control of cataract, Keshan disease and the Kaschin-Beck disease, delay senility and so on. Simultaneously selenium also detoxicats some heavy metal elements (for example mercury, arsenic, silver etc).Therefore, development into some medicine and health products containing selenium now becomes a research hot in medicines. The organic selenium compound is highly absorbed by people, strong biological activity, low toxicity, light environmental pollution and so on. It has become a present research direction of selenium. The research on organic selenium compound is on the rise in the domestic and foreign.According to the synthetic principle of environmental organic selenium compound, the research progress on organic selenium compound in the domestic and foreign was reviewed, The tea polyphenol with the biological activity ingredient was withdrew from Shaanxi' green dust tea. The structure was characterized by infrared spectrum, ultraviolet spectrum, high efficiency liquid chromatogram etc. It was established that a new method of preparation of tea polyphenol using the Shaanxi' green dust tea with the extracting agent of ethyl alcohol in Soxhlet extractor. Polyamide silica gel was used as absorbent for column chromatogram. The optimum process of the method was discussed. The optimum process: extraction agent of 70% ethyl alcohol solution, extracting with Soxhlet extractor for 1.5 hours before entering column chromatography. The column chromatogram is as follows: the column size (the altitude: diameter) is 15:1, the absorbent is 1: 1 of polyamide and silica gel, the granularity is 100 - 200 item, elution agent is 70% ethyl alcohol solution. In the optimum condition, the content of teapolyphenol is 94.5%, overall yield is 18.9%.From the view of pollution-free chemistry, using tea polyphenol from Shaanxi' green dust tea as raw materials, the synthetic route of selenylation tea polyphenol was obtained,it began at grignard reagent ,and the preparation of diaryl-diselenide compound then halogeno-aromatic selenium .And also anti-cancer character was discussed .The optimum response conditions:(l)The preparation of grignard reagent: solvent of ether, raw material of magnesium power, the mole ratio of ether and bromobenzene is 2:1, without initiator, the reaction time for 4 hours, yield of the products reaches above 90%.(2)The preparation of halogeno-aromatic selenium: the mole of selenium and the grignard reagent is 1:1, the mole ratio of liquid bromine and selenium is 2:1, the reaction temperature of 60'C, the reaction time for 4 hours, the yield of halogeno-aromatic selenium of 90.2%.(3)The preparation of the selenylation tea polyphenol: tea polyphenol was jioned before halogeno-aromatic selenium, the mass ratio of the tea polyphenol and aromatic halogeno-selenium is 3:10, the reaction temperature of 80X?, the reaction time for 5 hours, the yield is 91.3%, the content of selenium is 3.8 ug/g.It has been proved that the selenylation tea polyphenol has obvious toxicyst function to the cancer cell such as Hela, A549, HepG2. Suppress curve to the growth of the cancer cell was the standard S and the obvious dosage dependence. When dosage of tea polyphenol selenized is 300 mg/L,the restrain rate to Hela, HepG2 was above 60%, and highly 79.45% to A549, and greater than antineoplastic Fluorine urine pyrimidine, through t experiment. The suppress density to half is respectively 273.37, 171.46, 407.89 mg/L to Hela, A549 and HepG2 cell.
