The goal of this paper was the study on tat mediated magnetic nanoparticleshaving composite targeting function. Starch magnetic nanoparticlces (SMNPs) andO-carboxymethylated chitosan magnetic nanoparticles (OMNPs) with the averagediameter (<100nm) and super-paramagnetic property were obtained by the method ofco-precipitation with polymer matrix, respectively. In order to investigate thedrug-carrying capacity of the MNPs, MTX-SMNPs and MTX-OMNPs were preparedwith the method of EDCI. All the above magnetic nanoparticles were characterizedby TEM (JEOL 100CX-II), PSA (BI-90plus), AFM (Nanoscope IIIa, DI, USA), IRspectrum (Bio-Rad FTS 135) and VSM (Vibrating Samples Magnetometer,LDJ9600-1), which proved that the two MTX-MNPs preserved thesuper-paramagnetic property, the diameters were both below 100nm and the drugcontent of the two MTX-MNPs were 45.6%,41.088% respectively. In addition, in order to improve the targeting and translocational capability of theMTX-MNPs, we subsequently prepared MTX-TAT-MNPs, MTX-TF-MNPs andMTX-TAT-TF-MNPs. The morphology, average size / size distribution of all theabove drug delivery systems were measured by TEM, AFM, PSA and VSM. The resultsshowed that the average size of all the above carriers are below 100nm and still retainedwell-shaped morphology with narrow size distribution and magnetic properties that areclose to suerparamagnetic matters. HPLC-UV detection proved that TAT and TF werealmost completely combined covalently to the surface of the drug carrier system.Then C6 cell line was selected to investigate the anti-tumor effects of the BlankMNPs, MTX-MNPs, MTX-TAT-MNPs, MTX-TF-MNPs and MTX-TAT-TF-MNPs.The results demonstrated that all the MTX-loaded MNPs had high anti-tumor effects(>80%). The antitumor effects of the MTX-TAT-MNPs, MTX-TF-MNPs andMTX-TAT-TF-MNPs are much higher than that of the MTX-MNPs, especiallyMTX-TAT-TF-MNPs had the highest antitumor effect among all the MTX-loadedMNPs. The antitumor test also demonstrated that magnetic field applied to the MNPscould significantly improve the anti-tumor effects correspondently. In summary, TAT,TF and the magnetic field could significantly enhance the anti-tumor effects of theMTX-MNPs.
