The preparation methods of the powder phospholipids with high content acetone insoluble and phosphatidylcholine(PC)were obtained using Condensed Soybean Phospholipids(CSP) as raw materials, respectively. The main contents in this thesis are:The powder phospholipids with high content Acetone Insoluble(AI%) was obtained using anhydrous Acetone as solvents, and ultrasonic assistant extraction technique. The ultrasonic power, run time, and the extraction parameters were studied. The result showed that extraction temperature, times, ration of CSP to solvent, and rate of shear are the main factors to AI%. The optimal experiment conditions were achieved by orthogonal experiment. Under the condition of ultrasonic power 80w run 5min, optimal parameters of extracted five times, extraction temperature is 0℃, ratio of CSP to solvent is l/12g/ml, rate of shear is 5000 r/min, the high powder phospholipids with 98% Acetone Insoluble content was obtained. Ultrasonic assistant extraction technique and traditional extraction technique were contrasted. The result showed that AI% of ultrasonic assistant extraction technique was higher than traditional extraction technique about 3.3% averagely, and extraction time was shortening obviously.The ethanol was selected as extraction agent on the basis of organic and inorganic value ration (I/O) of phospholipids and low carbon alcohol. The feasibility of using CSP as raw materials, and aqueous ethanol as organic solvents was confirmed by testing CSP and powder phospholipids with high AI%. The effects of ethanol concentration, extraction temperature, ratio of CSP to solvent, and extraction time on PC purity and yield have been studied. The result showed that under the condition of ethanol concentration is 95% (v/v), extraction temperature is 30°C, ratio of CSP to solvent is 1:5-1:9g/ml, and extraction time is 30min, CSP can be made into powder phospholipids which PC purity can reach 50% upwards.Based on the first-step extraction with ethanol as solvent, a combination of second extraction and high-low temperature alternate treatment was examined. The effects of them on PC purity were investigated. The optimal experiment conditions were achieved by orthogonal experiment. It was shown that the low temperature is the most remarkable factor to PC purity. Under the optimal parameters of high temperature extraction times is 2, temperature is 30℃, and extraction time is 30min, low temperature is -20℃, extraction time is 20min, ratio of CSP to solvent is 1:5 g/ml, the PC content about 90% of phospholipids could be obtained using 95% (v/v) ethanol as solvent.Soybean phospholipids were analyzed by reversed phase-high performance liquid chromatography using gradient eluted program, a VWD ultraviolet detector, and acetonitrile/water as mobile phase.
