The situation of selling well in the world of traditional Chinese medicine (TCM) has accelerated the modernization process of traditional Chinese medicine, and the research of effective composition is the modernized key to TCM. Because microwave, supersonic technical and preparative liquid chromatography technology are fast and highly effective, they are focused attention on the preparative separation process of the active component in TCM. And the high-performance liquid chromatography (HPLC) - mass spectrum (MS) technology unified the highly effective separation potency of LC and the powerful analysis ability of MS. The technology has become one of the most powerful analysis methods in analysis area. This article has mainly studied the microwave, supersonic and the preparation liquid chromatography technology in extraction of active components in gingko, lotus leaf and the zizyphi spinosi semen (ZSS), and a HPLC coupled with UV photodiode-array detection and electrospray mass spectrometry (HPLC-DAD-ESI-MS) was developed for the simultaneous analysis and identification of main bioactive constituents in extract of zizyphi spinosi semen. The main content is as follows:1. To establish an isolation method of nuciferine by preparative HPLC. By contrast with the literature and standard reference substance, nuciferine obtained by this method was identified by MS, IR, UV, ~lHNMR, HPLC. The purityof product is higher than 98% and nuciferine yield from extracts by this method exceeds 49%. The developed method is simple, rapid and at low production cost. The product owns the quality of reference substance.2. A rapid microwave-assisted hydrolysis (MAH) method is presented for determining ginkgo flavonol glycosides in extracts of Ginkgo biloba L. (EGb). Three flavonol aglycones such as quercetin, kaempferol and isorhamnetin were analyzed by HPLC. The effects of solvents, acidity, power and time of microwave radiation have been studied on microwave-assisted hydrolysis. Comparing with traditional reflux hydrolysis (TRH), microwave-assisted hydrolysis method owns advantages such as faster, lower-cost.3. A HPLODAD-ESI-MS method was developed for the simultaneous analysis and identification of main bioactive constituents in ZSS and its extract ZSS, including three f lavonoids, three cyclopetide alkaloids and four triterpene saponins. This method can be quality control of ZSS and its extract.4. To optimize the extraction method of zizyphi spinosi semen extract. The effects of different concentration of different solvents have been studied on extraction of ZSS, and the process of extraction has been ameliorated. The new method is lower-cost and high-effective.
