Heparin (Hep) is a natural anticoagulant with an average molecular weightof about 15000. It consists of repeating disaccharide units of uronic/glucuronicacid and glucosamine residues. Owing to the dissociation of acid groups, thewhole heparin molecule is negatively charged in water solution and the averagecharge is ¨C70.It is a parenteral drug with a very rapid onset of action dueto its inhibition of clotting factors near the end of the coagulation cascade.It has been widely used in many clinical procedures for more than 60 years andnow it is still the first-choice to prevent thromboses and to cure urgent veinthrombus.Heparin and its derivants have a variety of biological activities suchas anticoagulant, antilipemic, antithrombotic, immunoregulatory,antiphlogistic and antianaphylactic activities, etc, So the heparin level inthe patient?ˉs blood needs to be carefully and accurately monitored duringsurgery and recovery.Adenosine disodium triphosphate (ATP) is the mediator of energy exchanges,which occur in all living cells, both catabolic, degradative processes andanabolic, biosynthesis processes. Because of its ubiquitous presence in livingmatter, ATP has been widely used as an index for biomass determinations inclinical microbiology, food quality control and environmental analyses.The nicotinamide adenine dinucleotide phosphate (NAD and NADP) are naturalcoenzymes present in all animal and plant organisms. They are essential to thefunction of many catabolic and biosynthetic pathways found within livingorganisms. Each of these forms has a particular role in enzyme regulation, duringdifferentiation and similarity processes.Nicotinamide adenine dinucleotidephosphate has a pyridine circle of nicotinamide, and using it oxygenation anddeoxidization can occur reversibly, it can be used for the transfer of hydrogenin the metabolizability. One of the appreciated roles of the nicotinamide adeninedinucleotide phosphate is in the prevention and/or reversal of oxidant damagewithin the cell. Therefore, it is significant to establish a precise, sensitiveand simple method for the determination of NADP.In this paper, according to above three objects, using molecule spectroscopy(fluorescence and Uv-Vis spectroscopy), we study the interaction betweendrug-metal ion complex and drug and its application. There are three chapters.First, using Tb3+-carbostyril complex as fluorecence probe, studied theinteraction between the complex and Heparin (Hep) and its application. By theRosenthanl graphic method, the association constant and binding numbers ofheparin with the probe were obtained.After addition Heparin to Tb 3+-carbostyrilcomplex system, the characteristic fluorescence intensity of Tb 3+ could beenhanced greatly, and the enhanced fluorescence intensity is in proportion tothe concentration of Heparin. According to this, we constructed a new methodfor the determination of Heparin. The developed method was practical, simple,sensitive and relative free from interference coexisting substances and had beensuccessfully applied to the determination of Heparin in biological samplesSecond, using Tb3+-carbostyril complex as fluorescence probe, studied theinteraction between the complex and adenosine disodium triphosphate (ATP) andits application. The carbostyril including norfloxacin and enxrofloxacin.Carbostyril is one kind of bacteriophages containing |á-carbonyl carboxylic acidconfiguration. Literature survey shows that |á-carbonyl carboxylic acidconfiguration is suitable for efficient energy transfer from ligand to Tb3+ ion,Therefore, carbostyril is ideal ligand for Tb3+ ion and it can possibly sensitizethe fluorescence intensity of Tb3+ ion via intramolecular energy transfer. AddingATP to system, the characteristic fluorescence intensity of Tb3+ could be enhancedgreatly, and the enhanced fluorescence intensity is in proportion to theconcentration of ATP. According to this, we constructed a new method for thedetermination of ATP and successfully applied to determination of ATP in samples.Moreover, the enhancement mechanisms of the fluorescence intensity in thecarbostyril -Tb3+ system, and the carbostyril -Tb3+-ATP system have been alsodiscussed.Third, there is energy transfer between ENX and Tb3+ and the complex can emitthe characteristic fluorescence of Tb3+, so the complex can be used to determineNADP as a kind of fluorescent probe. In addition, NADP can form a very stableternary complex with the ENX-Tb3+ system and can dramatically increase thefluorescent intensity of complex. In this part, we studied the fluorescenceincreased effect of NADP on the complex system, explained the interactionmechanism and determined NADP in synthetic water samples successfully.
