A laboratory experiment was conducted to evaluate the influence ofaccompanying anions on toxicity of lead in Vicia faba seedlings root using test theactivities of plasma membrane Ca2+-ATPase (PM—Ca2+-ATPase). Lead withdifferent accompanying anions was carried out, they were lead chloride (PbC12),lead acetate (Pb(OAc)2) and lead nitrate (Pb(NO3)2) respectively. The Pbconcentrations were 0, 1×10-7, 1×10-6, 1×10-5, 1×10-4, 1×10-3 mol/L in solutionswhich used to expose Vicia faba seedlings root to lead respectively.And theexposedtimewas12,24,36hoursrespectively.A standard protein curve and a standard phosphorus curve suitable for thisexperiment were worked out. Effect of Ca2+, ATP, Temperature, and pH on theactivityofPM—Ca2+-ATPasefromViciafabaseedlingsrootweredetermined.Low-temperature high-speed centrifuge Beckman J2-21 (rotor: JA-21) andaqueous two-phase extraction (ATPE) were used to obtain highly purified PMvesicles. The PM—localized Ca2+-ATPase activity was tested. And the influence ofaccompanying anions on activities of PM—Ca2+-ATPase from lead stressed Viciafabaseedlings rootwererespectivelytested.The result of activity of PM—Ca2+-ATPase from Vicia faba seedlings roottesting demonstrated that, the activity of PM—Ca2+-ATPase reached the maximumwhileCa2+concentrationwas3mmol/LandATPconcentrationwas3mmol/L...
