| A study on the extraction of capsanthin and capsaicin by using supercritical fluid technique in capsicum was carried out. It is of great value to accelerate the development of the economy, and it can also increase the annual income of the farmers.In this thesis the capsanthin and capsaicin were prepared from pepper by supercritical fluid extraction. Many factors such as extraction pressure, extraction temperature, separation pressure, separation temperature, fluid flux were used to estimate the color value and yield of capsanthin, the content and yield of capsaicin. The capsanthin's color value and other capability of stability (such as the stability effect of light, heat, acid, alkali, metallic, oxidizer and reducer) were studied. In order to improve the stability of capsanthin, the addition of pepper seed extraction was also carried out.The results showed that:(1) The experiment detected the color value and yield index of capsanthin extracted with supercritical CO2 fluid extraction and solvent extraction. The result illustrated that supercritical CO2 fluid extraction was better than solvent extraction, and the supercritical CO2 fluid extraction's characteristics consist of the high purity, without remainder solvent, and so on.(2) The optimum conditions of capsanthin and capsaicin prepared by supercritical CO2 fluid extraction were determined by orthogonal experiments: the raw material size 40 80mm2, extraction pressure 20Mpa, extraction temperature 40℃, divide pressure 10Mpa, divide temperature 50℃, CO2 flux 20L·h-1, extraction time 3hs. The color value of capsanthin was 227.15, the yield of capsanthin was 9.54%, the content of capsaicin was 14.75%, the yield of capsaicin was 3.19%.(3) The effects of each factor to the color value and yield of capsanthin, the content and yield of capsaicin were studied: extraction pressure>extraction temperature>separation pressure>fluid flux.(4) Study on the stability of capsanthin with two methods showed that the capsanthin had a good resistance to light, heat, reducer, and it was suitable for neutral solution. Oxidizer, Fe2+, Cu2+ decreased the stability of capsanthin while Fe3+had little effect. |
