Dye-wastewater has five basic characteristics with high chroma,high COD,high salinity,high toxicity and low B/C,which is the "4 high & 1 low".The dye can steadily exist in the water for long. At present,about 100,000 varieties of dye are products in the world yearly,and the total annual production reaches about 700,000 tons.China is one of the main countries for dye production,in which 150,000 tons of dye are producted yearly.Effectual disposal for dye-wastewater is an urgent problem for China."Phanerochaete chrysosporium disposal" is a new technology for effectual dye-wastewater disposal.The special catabolic enzymes secreted by Phanerochaete chrysosporium is used with some other methods to degrade the synthetic dye into CO2 and H2O.This thesis continues the predecessor's work,and keep on doing the deeper research,explore the steady model that the open Phanerochaete chrysosporium bioreactor dispose the dye-wastewater, and use Molecule- Biology to study the dynamic change of the microbial community when the reactor is running and the relativity between the change and the reactor control and dye disposal.The main subjects in this thesis are:(1) How to build and run Phanerochaete chrysosporium bioreactor(2) Perfect the method to extract DNA(3) Do research on the microbial diversity during the dye decomposition with the PCR-DGGE method, and ascertain the dominant species.(4) Study the isozyme zymogram of biologic membrane specimen in different phases(5) Do research on The structure of biologic membrane in Phanerochaete chrysosporium with SEM.According the experimentation analysis,we can get the conclusion as follows:(1) Snailase is effectively for extracting DNA from fungi.(2) Phanerochaete chrysosporium is always the dominant species in the bioreactor,and carry on the leading role in the reactive brilliant red X-3B decompositionwhen the bioreactoris running.(3) Although it does not change the structure of gene ITS II obviously,it affects the expression of Phanerochaete chrysosporium Lip gene.(4) That the dye- waste water is disposed chronically and continuously will have a...
