Synthesis And Analysis Of Both Flavine Ademine Dinucleotide And Niclosamide

Flavin adenine dinucleotide(FAD) is an active coenzyme for yellow enzymes of phosphorylation derived from vitamin B₂ in vivo. It may also participate in metabolisms of carbohydrate, protein and fat for maintaining the normal visual function and promoting growth.. Vitamin B₂ deficiency disease may appear lack of FAD. Several methods have been proposed for chemical synthesise,instrument analysis and column chromatographies.Three techniques of chemical synthesize of FAD has been compared: The fiest method has obtained that purification of FAD is 70.1% and the yield is 25.6%; The second method has obtained that purification of FAD is 58.3% and the yield is 45.4%; The last method has obtained that purification of FAD is 55.5% and the yield is 35.2%. Primary product of FAD has been purified by a poly styrene porous polystyrene non-ion exchangeable resin , the phenol formaldehyde cation exchange resin and the phenol cation exchange resin. The factors of the column chromatographic have also optimized through the orthogonal experiment. The pretreatment of the resins , installing column and the column chromatographic have been searchedNiclosamide,as originally a agent to kill the oncomelania, is now used as the anti-parasite drug and for preventing schistosomiasis. It is also the only medicine of killing snails that was still recommendated by World Health Organization (WHO) since 1972. Here, some methods on chemical synthesise, the purification and the analysis of niclosamide has been examined . Primary product of niclosamide is condensated with 5-Chlorosalicylic acid, o-Chloro-p -nitroaniline and the catalyst of phase shift under no solvent system.The amine salt of niclosamide is synthesized with the primary product of niclosamide and organic amine. After filtered, neutralized, washed and dried, niclosamide for medicine is prepared. The factors has optimized through the orthogonal experiment. We have applied for Chinese Patents that is"A new method for preparation of niclosamide"(public number: CN1687016A) and"The prep aration of high purity and good yield niclosamide"(public number:CN 1626506A). With referrence of Chinese pharmacopoeia and British pharmacopoeia, the HPLC analysis method of niclosamide has been made.Flavin adenine dinucleotide disodium salt and niclosamide have been obtained. At the same time, they are affirmed by the element analysis , infrared absorption, mass, ultra-visible, ¹H- nuclear magnetic resonance and ¹³C- nuclear magnetic resonance spectra .
