Kinase, which catalyze the translocation of phosphate bond of ATP to the acceptor, are significant in the drug manufacture, the clinic use and the research on the anti-cancer drug. In this paper, self-controlled affinity chromatography materials were applied for the purification of the kinase extracted from the beer yeast. The technology of synthesis of the affinity chromatography materials and the affinity column chromatography were studied.The crude enzyme was extracted from the beer yeast through pretreatment and frost-lysis of the beer yeast cell. A 3.3-fold purified HK (Hexokinase, EC2.7.1.1) with a yield of 70.9% was obtained by collecting the precipitation between 30% ~70% ammonium sulfate.Based on the study on the selection of the ligand, etc, the process route of the synthesis of the affinity chromatography materials for kinase was ascertained, which include the carrier paper fiber, the activator apoxy...
